A total of 30 sites were established and monitored in 2006, 2007 and 2008. After the completion of 1-mile bridge in 2012, additional 10 new sites were established to observe the ecological impact of 1-mile bridge (known as Bridge & Census sites). In 2015, additional 40 sites were established along eight transects (T1-T8, known as near canal sites) in ENP marshes starting at, and roughly perpendicular to the L-29 canal. The purpose of these sites was to monitor the potential effects of Modified Water Deliveries (MWD) operations on changing nutrient concentrations and ratios in key ecological compartments due to increased downstream discharges from the L-29 canal beneath the 1-mile and 2.6-mile bridges and culverts along Tamiami Trail. area will be obtained from the Everglades Depth Estimation Network (EDEN) (http://sofia.usgs.gov/eden/) and will be used to calculate hydroperiod (inundation period) across the region. During site visits, water depth will be measured by standard rule (meter stick) to the nearest cm as the depth between the surface of the water and the resistance of the soil. This procedure was successfully used in prior studies (Scheidt and Kalla 2007; Gaiser et al. 2009; Sarker et al. 2020). These data will be used to characterize site-specific hydrologic conditions and also can be used to refine EDEN model estimates for future NESRS assessment. We measured hydroperiod as the number of days per year with water at or above a minimum depth threshold (Zedler 2000). Total wet and dry days for each site were summed for a particular year at each site to calculate hydroperiod (i.e., numbers of days per year with water depth >5 cm). We calculated hydroperiod from 01 May through 30 April based on daily water depth at each site for the water year (WY). Hydroperiod was calculated for a full WY starting from May to April of the following year, that is, hydroperiod for WY2024 was calculated from 1 May, 2023 to 30 April, 2024. Water Quality Water grab samples will be collected whenever sufficient surface water is present. Generally, water depth needs to be greater than 10 cm to conduct field measures of pH, and temperature using the Manta multiprobe (see below). Surface water grab samples were split into filtered and unfiltered subsamples according to the following protocol. Approximately 500 mL of surface water was collected from mid-depth in the water column (lower boundary floc/water interface, upper boundary water/air interface) at each location. Surface water was collected in a 500 mL Nalgene bottle (rinsed 3 times at site) covered with 100 µm Nytex screening to exclude floating debris. This sample was then split with approximately 250 mL being used to rinse and fill a 140 mL syringe to which a 0.45 µm membrane filter-containing filter holder is attached. A small portion of filtered water was used to rinse (3 times) a 60 mL high-density polyethylene (HDPE) sample bottle. This bottle was then filled with filtered water. Analysis and QA/QC was performed according to standard operating procedures determined by ENP and FIU. Generally, all surface water samples were delivered from the field to the respective lab on ice and will then be refrigerated if not analyzed immediately (< 24 h). Filtered and unfiltered samples were analyzed using standard protocols. At each sampling location a Manta+30 multiprobe will be used to measure dissolved oxygen (DO), temperature, pH, and specific conductance at the mid-point of the water column, using a standard ruler to measure the water depth from the air/water interface to the soil surface (by resistance). Water Analytical Methods as used in FIU Laboratories: Samples were analyzed using standard analytical methods. Water was analyzed for soluble reactive orthophosphate (SRP; USEPA method 365.1), and total phosphorus (TP; USEPA 365.1 following dry ashing according to Solorzano and Sharp 1980), soluble nitrate, nitrite (NO3, NO2; USEPA 353.2) and ammonium (NH4; USEPA 350.1) on an automated colorimeter. Total nitrogen (TN: ASTM D5176) and total organic C, if applicable (TOC; USEPA 415.1) were determined on a Shimadzu TOC-VCSH fitted with a Shimadzu TNM-1 Total Nitrogen Analyzer. Soil and Flocculent Detrital Organic Matter (Floc): A common feature of Everglades long-hydroperiod wetlands is the presence of a layer of flocculent detrital organic matter (floc) overlying the soil surface. It is probable that changes brought about by increased water flow will affect the physiochemistry of this layer before causing changes in the surficial soils (SS; 0-2 cm) (Gaiser et al. 2005) and surficial soils should show changes before deeper soils (DS; 2-10 cm). We sampled, by intact coring, the soil (SS and DS) and the overlying floc at all sites according to the schedule. We define floc as an “unconsolidated” layer meaning that it lacks the coherence of soil and therefore “flows” off the soil when extruded from a core. Three replicate samples of soils and floc were collected from each site at locations determined by the over-the-back tossing of the periphyton quadrant (see below). That is, within a site, soil/floc sampling occurred at the same locations and prior to the periphyton collection. Floc depth was determined by visual measurement of a standard rule as the distance between the soil surface (by resistance) and the bottom of the water column. Additionally, depth will be verified within collected cores where possible. Cores were collected by inserting a plastic tube of known diameter (surface area) through the floc and into the soil. Care will be taken to minimize compaction. If compaction was observed then cores were collected as individual increments. Once collected, the cores were separated into the floc (single measured depth increment) and the 0-2 cm soil depth increment. Generally, the smallest diameter core was used to collect soil and floc. The size of the core tube used depended on soil/floc conditions and was determined by the ability to collect sufficient material for all analysis in as few core insertions as possible. Core tubes were of two sizes; a small diameter core (2.35 cm in diameter) and a larger core (5.10 cm in diameter). Where possible, a single core was collected from each of the three replicate quadrants. Soil and floc samples were put on ice in a cooler and transported to the appropriate laboratory depending on site type and analysis needed. Soil and Floc Handling and Analytical Methods as used in FIU Laboratories: Soil and floc samples were put on ice and returned to the laboratory where they were homogenized and large anomalous material (white roots, snail shells, etc.) were removed. The soil and floc samples were processed for dry weight (40°C), field bulk densities (g dry weight cm-3), and ash-free dry matter Loss-on-Ignition LOI) at FIU. The dried and ground soil and floc were analyzed for TP, TN, and TC according to standard protocols for solid materials and tissues. Samples were analyzed using standard analytical methods. Soil and floc samples were weighed for total wet and dry weights (after drying at 40º C until constant weight) to determine bulk density and percent moisture. Subsamples were combusted in a muffle furnace at 550°C for 3 h to obtain ash content and organic matter by loss on ignition (LOI; Nelson and Sommers, 1996). Plant tissues and periphyton samples were processed using similar analysis for TP, TC, and TN if applicable. Total phosphorus in soil, floc, or tissues was determined using the ashing/acid hydrolysis method of Solorzano and Sharp (1980) with the resulting soluble reactive phosphorus (SRP) being measured as above. Solid samples were analyzed for total C and N using a Perkin Elmer Series II 2400 CHNS/O Analyzer (Nelson and Sommers, 1996). Periphyton When present, a 120-mL subsample of periphyton from triplicate samples was collected in a sterile bag, placed on ice, and analyzed in the laboratory for dry mass, ash-free dry mass, chlorophyll a content (a measure of algal abundance), organic content, and total carbon, nitrogen, and phosphorus (TC, TN, and TP, respectively) concentrations using the methods described above for soils and floc unless where noted below. When periphyton was absent, the surface floc layer containing algae was sampled to assess organic, chlorophyll a, and nutrient content. All field data were doubly entered into spreadsheets for quality control screening. Periphyton was collected and analyzed using standard protocols approved by the U.S. EPA and published for Everglades periphyton (Stevenson & Bahls 1999; Gaiser et al. 2004, 2005, 2006). Periphyton was homogenized in a measured volume of distilled water. We removed volumetric subsamples for analysis of dry weight, ash-free dry weight and total phosphorus. Dry weight was determined after samples have reached constant weight in a 100ºC oven. Ash-free dry weight was measured after combusting the dried samples at 500ºC for 1 h. Total P will be measured on dried material using standard colorimetric methods (see above for floc and soils). Chlorophyll a was measured fluorometrically to estimate algal biomass on subsamples of fresh materials. All subsample estimates of periphyton abundance were scaled-up to plot (m2) level using the plot-specific periphyton biovolume measured in the field. Samples submitted to the FIU lab for nutrient analysis were dried and ground in the periphyton laboratory. All samples have been processed and data doubly entered into spreadsheets for quality control screening. Vegetation Nutrient Analysis A sample of Cladium jamaicense tissue was collected from the central 15 to 20 cm of one leaf of three plants at or near each of the quadrats used for soil, floc, and periphyton sampling. The three leaves were composited to provide one sample per quad, three replicates per site. This methodology for the tissue collection and laboratory analysis follows the protocols applied in the Gaiser et al. (2009) monitoring including drying, grinding and submission to the FIU lab for nutrient analysis. Laboratory Quality Assurance / Quality Control (QA/QC): Florida International University maintains strict Quality Assurance and Quality Control procedures. Calibration curves are analyzed and summarized at the beginning of all analytical runs according to method specification. Standards, either citrus or apple leaves, are digested or combusted for all TP and TN/TC analysis. All standards are accepted within the analytical range as defined by the National Bureau of Standards (NBS). Analytical duplicate samples are analyzed (10% of total samples) in all analysis runs. Acceptable duplicate analysis is ± 15% relative percent difference (RPD). Matrix spikes are analyzed at 5% of total samples in all analytical runs and are accepted at ± 20% recovery. Additionally, analytical blanks are (5% of total samples) analyzed in all runs. All QA/QC results are included with combined data file. The Southeast Environmental Research Center within the Institute of Environment at FIU utilized Chain of Custody forms to ensure all samples and analysis are accounted for and tracked.
