Glass slides were used as artificial substrates for periphyton growth. Twenty slides were incubated for 2 months in triplicate periphytometers (flow-through plastic boxes) at the surface (marsh sites) and bottom (seagrass sites) at each site. In addition, at seagrass sites, twelve artificial seagrass blades were incubated for two months in triplicate at each site. Blades were made from strips of transparency film anchored to the bottom and floating upward toward the surface by use of a styrofoam bead on the top of the blade. Data are presented in terms of grams dry and ash-free dry mass accumulated per area per time, as well as the chlorophyll a content of the dry mass and its rate of accumulation. Total phosphorus content of accumulated periphyton is also provided. This is part of continuous data collection to test the hypothesis that phosphorus availability and hydroperiod influence periphyton production in the Everglades. Triplicate periphytometers, each containing 20 slides, were incubated at the water surface (SRS sites) and on top of the sediments (FB sites); artificial seagrass blades were incubated at Florida Bay sites. Periphytometers were placed near the markers for each site. Method Step Description Periphytometers and artificial blades were collected after 2 months of incubation and returned on ice to the lab. Periphyton was scraped from slides into beakers and homogenized in distilled water. They are then entered into the appropriate digital format. These data are reviewed in hard copy and digital format by the lab manager. Laboratory data are written in pencil on waterproof paper, photocopied prior to computer entry and then entered digitally in duplicate and cross-referenced to hard copy notes. Data outliers, determined by comparison to long-term variance in that parameter, are highlighted in resulting spreadsheets for examination by the principal investigator, who may discard the values. Information Management Notes Information Manager (December 19, 2011) 1) The file LT_PP_Gaiser_003 should NOT include %n and %C and SHOULD include the Organic Content (ORG) column. This new file replaces the old data files (ST_PP_Gaiser_001 & all versions) so that these values are all updated. Dr. Gaiser decided to drop the N and C because they often don't have enough material to reliably analyze that, and the Organic Content shows interesting patterns that are important to report, 2) Changed date format to DD-MMM-YY and coded missing values with -9999 (including changing NES and NS to -9999), 3) Reordered and renamed columns to SITENAME, Date, Substrate, Replicate, IncubationTime, DryWeight, AFDM, ORG and TP, 4) Corrected site names to reflect TS/Ph instead of TSPH.
