We quantify metabolic rates of periphyton assemblages using standard oxygen change techniques in 300mL light and dark BOD bottles in triplicate. Using YSI dissolved oxygen probes and meters, we measure dissolved oxygen change as the difference of initial and final oxygen concentrations during a two hour incubation period. We calculate net primary productivity and respiration rates in units of oxygen and carbon, then normalize to the organic content (ash free dry weight) of the incubated periphyton. We quantify metabolic rates of periphyton assemblages using 12 300mL light/dark BOD bottles per site. Ambient water is placed into six light and six dark BOD bottles. Then approxiamately 500 mm2 of periphyton is added into three light and three dark bottles, the remaining six bottles are controls. Using YSI dissolved oxygen probes and meters, initial and final dissolved oxygen concentrations and times for all bottles is measured during incubation (approxiametely two hours). Incubated periphyton is brought to the lab and dried at 70 C for a minimum of 24 hours then ashed at 500 C for 3 hours. We calculate net primary productivity and respiration rates using standard oxygen change techniques to units of carbon, then normalize to the organic content (using the difference of dry weight and ash free dry weight, AFDW) of the incubated periphyton.
