MethodsSampling Description
The sites TS/Ph 7a, 8, 9, 10 and 11 were chosen as the main LTER sampling sites in Florida Bay, because they were sites where a lot of historical data were collected and they also are representative of the salinity gradient across Florida Bay. We are assessing total carbon, nitrogen, and phosphorus at these sites because of the importance of the makeup of the ecological communities at those sites.

Description
At each sampling site, 5 intact short shoots of Thalassia testudinum (and a minimum of 10 Syringodium filiforme and 15 Halodule wrightii short shoots, if present) were haphazardly collected from a 10 m2 area. These short shoots were returned to the lab, where all attached green leaves were cut from the short shoots and cleaned of adhering epiphytes by gently scraping with a razor blade. All leaves from a site were pooled and dried at 80 degrees C. Dried leaves were ground to a fine powder using a ceramic mortar and pestle. Powdered samples were analyzed in duplicate for carbon and nitrogen content using a CHN analyzer (Fisons NA1500). Phosphorus content was determined by a dry-oxidation, acid hydrolysis extraction followed by a colorimetric analysis of phosphate concentration of the extract (Fourqurean et al.1992a). Elemental content was calculated on a dry weight basis; elemental ratios were calculated on a mole:mole basis.
